Description
Requirements:
Provides accurate dynamic control of temperature, humidity, CO2 and oxygen to maintain required cell culture conditions for live cell confocal and STED super-resolution imaging. The stage top incubator is required in order to perform live cell imaging with the unique resolution capabilities of our Leica Stellaris 5 STED microscope. It currently does not have a stage top incubator, and therefore, is unable to perform this type of imaging requested by NCI researchers.
Stage top incubator controller must have two feedback modes, chamber temperature or specimen temperature, while it also continuously monitors the room temperature to maintain specimen temperature within set point anywhere between 25 – 50 degrees Celsius. The built-in digital gas mixer controller accepts 100% CO2 from lab supply to regulate CO2 concentration at the sample, with a CO2 range of 5.0 to 20% with real-time sample feedback. In addition, humidity is actively controlled with an internal humidifier to sense sample chamber humidity. The electrically heated stage top incubator chamber must have magnets for easy interchange of specimen holders, including inserts for 35 mm coverglass dish, 50/60mm coverglass dish, chamber slide, coverglass chamber, and open frame insert for imaging multi-well chambers. A flexible heated collar that accommodates a range of objective lens diameters must also be included.
