Description
Agonist-activated PI3K signaling and nuclear phosphoinositide (PI) signaling in response to stress have fundamental roles in many cellular functions and diseases including neurodegenerative disorders and cancer. The research aims to reveal mechanisms for spatial PI signaling in the cytosol and nucleus, with broad implications for cancer, neurodegenerative disorders, and other diseases. The project investigates the regulation of PI3K signaling on cytoplasmic membranes via scaffolding mechanisms, the assembly of PI3K complexes at membranes and endosomes, and the linkage of PIPn to nuclear proteins such as p53, which influences tumor suppressor and oncogenic functions. The study also explores the chemical nature and enzymatic mechanisms of PIPn linkage, the cellular PIPylome, and the role of PIPylation in DNA repair.
