Description
This project aims to utilize genomic approaches and targeted genome editing (CRISPR/Cas9) to evaluate the role of genetic diversity in malaria vaccine candidate antigens, including PfRh5 and its binding partners. By creating transgenic parasites with specific variants, the study will assess immune evasion mechanisms and vaccine efficacy prior to Phase 2 clinical trials. The research will involve analyzing IgG from malaria-immune individuals and vaccine trial participants through growth inhibition assays, neutrophil respiratory burst, and opsonophagocytosis tests. The project emphasizes integrating genomic sequencing, structural biology, and standardized immunological assays, leveraging collaborations with endemic countries. The goal is to identify vaccine candidates capable of eliciting broadly neutralizing antibodies, thereby advancing malaria vaccine development and overcoming parasite immune evasion strategies.
