Description

This project investigates how the ubiquitin-mediated degradation of the retinoblastoma (Rb) tumor suppressor controls cell cycle progression and therapeutic response in breast cancer. The research aims to identify the ubiquitin ligase (E3) responsible for Rb degradation and to understand how Rb degradation affects cell cycle regulation and response to Cdk4/6 inhibitors. The study involves mass spectrometry, CRISPR/Cas9 gene editing, and live-cell imaging to elucidate the timing and regulation of Rb degradation. The findings could provide insights into overcoming resistance to Cdk4/6 inhibitors and enhance understanding of Rb’s role beyond phosphorylation-dependent inactivation.
