This NIH grant funds structural studies of the eukaryotic transcription Mediator complex using biochemical and cryo-EM methods to understand gene expression regulation mechanisms relevant to development and disease.

Description

The project aims to use biochemistry, genetics, macromolecular electron microscopy, and advanced image analysis techniques to discover how Mediator performs its essential function in controlling gene expression. The results are critical to public health because dysregulation of gene expression is directly connected to developmental disorders, cancer, and other human pathologies. The studies will utilize cryo-EM, image analysis, and biochemistry to understand how signals from transcription factor binding to various mammalian Mediator subunits converge to control interaction with RNA polymerase II.
