Description

This project aims to develop high-throughput, multiplexable assays to systematically characterize how SAVI mutations affect STING-mediated transcription, autophagy, and cell death across common human genetic backgrounds. The work includes creating a barcoded library of SAVI mutations in cis with the five most common human STING alleles, using synthetic transcriptional reporters for IRF3, NF-κB, and TFEB/TFE3 activity, and evaluating autophagy capacity via FACS-based GFP-LC3 puncta formation assay. The goal is to understand how variants modulate STING’s functions and to inform targeted therapeutic strategies for STING-related disorders.
