Description
The Immune Data Science (IDS) Core will analyze high-dimensional immune datasets generated by Projects 1-3 and the Model Systems (MODS) Core. The analysis aims to address viral factors contributing to transmission, immune factors blocking transmission, efficacy of passive immunization and vaccines, immunogenicity, and biomarkers. Data categories include multiplex cytokines, antibody concentrations, flow cytometry, scRNA-seq, antigen receptor repertoire, and spatial proteomics/transcriptomics. The primary outcome is viral transmission, defined as detectable CMV by qPCR. Additional outcomes include maternal plasma viral load, shedding in urine and saliva, and dissemination across tissues. The first aim (SA1) will develop computational pipelines for processing, quality control, and exploratory analysis of immune data, focusing on single-cell data, antigen receptor diversity, and spatial biology. The second aim (SA2) will develop statistical models to link data, context, and outcomes, including hierarchical Bayesian models and spatial co-localization analyses. Overall, the IDS Core will serve as the quantitative hub, providing expertise and resources for complex data analysis and interpretation.
