Development of an injectable biodegradable hydrogel depot for single-dose self-replicating mRNA vaccine delivery targeting Staphylococcus aureus infections to improve immunization efficacy and patient compliance.

Description

This R01 will develop an injectable vaccine depot comprising cationic polymers to condense and charge neutralize anionic self-replicating mRNA vaccines into nanometer-sized particles incorporated within an injectable biodegradable gel of N-succinyl-chitosan and oxidized alginate. The project aims to generate a self-immunizing biomaterials technology superior to repeated systemic bolus injections.
