Description
The project aims to develop new technologies for single-cell assessment of DNA methylation, enabling high-throughput, cost-effective, and precise analysis across development and disease research disciplines. It proposes to leverage cutting-edge cell encapsulation, transposase-based library construction, and nanopore sequencing to develop a platform for direct detection of DNA methylation in previously inaccessible genomic regions, including repetitive regions and allele-specific methylation patterns. The technologies will be demonstrated on diverse samples including human PBMCs, cerebral cortex, induced pluripotent stem cells, and control cell lines.
