This NIH-funded project at Temple University aims to map human chaperone networks in yeast and human cell models to identify therapeutic targets for neurodegenerative diseases associated with protein aggregation.

Description

The project aims to build the first exhaustive map of a chaperone network against ADRD-associated proteins TDP43, FUS, and TAU. Using a novel genetic technique, all 12,155 possible combinations of the human Hsp40-Hsp70-Hsp110 network will be expressed and studied in yeast models. The active chaperones will be further analyzed in human cells and in vitro assays to assess their ability to prevent and reverse TDP43 aggregation. The project will also expand to study TAU aggregation and develop a system for overexpressing all 194 human chaperones in human cell models to identify therapeutic targets for ADRD.
